ABSTRACT

italic>Fusarium oxysporum widely exists in farmland soil and is one of the main pathogenic fungi of root rot, which seriously affects the growth and development of plants and often causes serious losses of cash crops. In order to screen out natural compounds that inhibit the activity of Fusarium oxysporum more economically and efficiently, random forest, support vector machine and artificial neural network based on machine learning algorithms were constructed using the information of known inhibitory compounds in ChEMBL database in this study. And the antibacterial activity of the screened drugs was verified thereafter. The results showed that the prediction accuracy of the three models reached 77.58%, 83.03% and 81.21%, respectively. Based on the inhibition experiment, the best inhibition effect (MIC = 0.312 5 mg·mL-1) of ononin was verified. The virtual screening method proposed in this study provides ideas for the development and creation of new pesticides derived from natural products, and the screened ononin is expected to be a potential lead compound for the development of novel inhibitors of Fusarium oxysporum.

ABSTRACT

Drug resistance resulting from bacterial biofilms can invalidate antibacterial agents. Therefore, eradicating bacterial biofilms to reverse drug resistance is a hotspot in the pharmaceutical research. In recent years, numerous studies have revealed the complicated mechanism of bacterial biofilm formation and strong drug resistance with multiple influential factors involved. This paper gives a comprehensive review on the process of biofilm formation and intervention by natural drugs, which can provide some reference and evidence for the following studies.

ABSTRACT

Ischemic heart disease (IHD), which has been considered to be exclusively caused by stenosis or occlusion of coronary artery, is a significant cause of morbidity and mortality worldwide. Mitochondrial dysfunction is the main pathological basis of ischemic heart disease and reperfusion injury, and moderate mitochondrial autophagy can selectively remove damage proteins and organelles to maintain intracellular homeostasis, so mitochondrial autophagy is important for maintaining the homeostasis of cardiomyocytes. Natural drugs from plants are widely used in ischemic heart disease. In recent years, more and more natural drugs have been proven to alleviate myocardial cell damage after ischemia/reperfusion through mitochondrial autophagy. This paper reviews the research progress of natural drugs from plants medicines regulating mitochondrial autophagy in the treatment of ischemia heart disease.

ABSTRACT

Natural lignans are a kind of plant secondary metabolites derived from the process of the oxidative polymerization of two molecules phenylpropanoids. Natural lignans have many pharmacological activities, such as anticancer, anti-inflammation, anti-virus, anti-oxidation, and liver protection. Using the methods of systems biology, biosynthetic pathways of lignans have been elucidated so far. By modifying the genetic expression of plants or microorganisms through metabolic engineering and synthetic biology, the synthesis of lignans and their derivatives have become more and more popular. The research progress in biosynthesis and engineering modification of lignans both in China and abroad is reviewed and discussed in this paper, with the purpose of throwing light upon the research and development of Chinese materia medica with lignans ingredients.

ABSTRACT

Objective To investigate epidemic characteristics of HCV and the distribution of naturally occurring resistance mutations of HCVNS5B in Hubei province, and provide the basis for individualized antiviral treatment for HCV infected pa-tients.Methods Collected HCV blood samples of 253 patients from March 2011 to May 2014 in Renming Hospital of Wu-han University, and determined the subtypes of HCVNS5B by first generation of sequencing technology, then compared the result with the nucleotide sequence of HCVNS5B online, located drug resistance loci and record. Results There were 4 gen-otypes and 6 different genetic subtypes in these 253 cases, namely 1a subtype 3,1b subtype 192,2a subtype 38,3a subtype 3, 3b subtypes 11 and 6a subtypes 6.Found 27 gene locus of resistance from 234 different places. The main drug resistance lo-ciare L159F (475-477)26.42%,followed by C316Y (946-948)16.98%,V321I (961-963)15.09% and S282T (844-846) 12.58%.Conclusion The epidemic characteristics of HCV in Hubei province were consistent with those of current in Chi-na;subtype 1b was the main types, followed by 2a subtype. The number of drug-resistant of HCVNS5B was more and com-plex,and presented some popular distributions. It was complex in mutations of 2a subtype, including 13 mutations of natural resistance. There were fewer mutations in subtype 1b,including sites of L159F and C316N.These results may provide the basis for antiviral therapy for HCV patients in this region.
